This project focuses on investigating novel molecular pathways responsible for resolving alternative non-B form DNA structures (such as hairpins, cruciforms, and G-quadruplexes) that can threaten genome integrity and are linked to genetic and neurodegenerative disorders. The research will employ the Xenopus egg extract system, defined DNA templates, biochemical assays, and mass spectrometry to dissect the unfolding mechanisms during DNA replication, aiming to uncover how these structures are detected, processed, and their implications for genome stability and human disease onset.
